Posto lungo la Via Amerina, Lugnano in Teverina fu fondato nel 600 d.C. dagli abitanti delle zone limitrofe per proteggersi dalle invasioni dei popoli barbarici prima, e dei saraceni in seguito.
Durante il medioevo, le città di Amelia, Todi e Orvieto si disputarono il dominio di Lugnano. Dopo aver vinto le altre città, Orvieto impose il proprio dominio su Lugnano per un lungo periodo. Lugnano in teverina oggi merita di essere visitata soprattutto per la magnifica chiesa di Santa Maria Assunta, gemma romanica, caratterizzata da un magnifico portico a colonnato. La chiesa si sviluppa su 3 livelli distinti, comprendenti la cripta e il coro.
A partire dal 1988, il paese è anche conosciuto grazie alla scoperta di una Villa Romana.
